PT-141 (Bremelanotide) is a melanocortin peptide that acts at the level of the central nervous system and binds to direct influence over sexual desire, arousal, and the neurological mechanisms of libido.

What is PT-141 (Bremelanotide)?

PT-141 is a synthetic peptide from the group of melanocortin receptor agonists, which primarily activates MC3 and MC4 receptors in the brain—key structures involved in regulating sexual behaviour, motivation, and arousal.

Unlike classic sexual function preparations that work peripherally by improving blood flow, PT-141 works centrally—directly on the brain mechanisms that control desire. This is a fundamental difference that positions it as a completely different type of molecule.

The peptide originates from research on melanocortin analogues, initially developed based on Melanotan II, but subsequently optimised so that it retains the effect on libido without a strong influence on pigmentation.

The active molecule bremelanotide was developed by Palatin Technologies and in 2019 was approved in the United States under the name Vyleesi for the treatment of hypoactive sexual desire in women, which further confirms its real action on central libido mechanisms.

What are the benefits of PT-141 (Bremelanotide)?

PT-141 is considered one of the most specifically targeted peptides for sexual function, with its effect not coming from physical stimulation, but from activating neurological pathways.

  • Increased sexual desire

The peptide activates the brain centres responsible for libido, which can lead to a clearly expressed increase in sexual desire.

  • Improved sexual arousal

It is associated with a stronger and faster response to sexual stimuli, by influencing the neuronal pathways of arousal.

  • Support for sexual function in men

In studies, an effect on erectile function has been observed, especially when the issue is related to a central rather than a vascular cause.

  • Improved sexual satisfaction

Some of the data indicate increased satisfaction and a more fulfilling sexual response.

  • A central mechanism of action

The main advantage of PT-141 is that it works at the level of the brain, not just the circulatory system, making it effective in cases where classic solutions do not produce results.

Clinical trials on PT-141 (Bremelanotide)

PT-141 is one of the few peptides in this category with real clinical data.

In clinical studies in women with HSDD, the following was observed:

  • increased sexual desire
  • increased frequency of satisfying sexual activity
  • improvement in the subjective assessment of sexual experience

In men, there is also data indicating improvement in sexual arousal and erectile response, especially in cases where the issue is related to neurological factors.

These results clearly show that PT-141 is not simply a peripheral stimulant, but a peptide with a direct effect on the brain’s regulation of sexual behaviour.

Correct intake and dosage of PT-141 (Bremelanotide) within a research protocol

In research protocols, PT-141 is typically administered in doses between 0.5 and 2 mg, used as needed, approximately 30 to 60 minutes before expected activity.

The approach is different from classic peptides, as it is not used daily but situation-based. Usually, a limited frequency of use is applied, avoiding excessive use in order to preserve receptor sensitivity and the stability of the effect.

Possible side effects of PT-141 (Bremelanotide)

As with most melanocortin peptides, unwanted reactions are primarily related to the central action of the molecule.

  • Most common side effects:
  • nausea
  • flushing of the face
  • headache
  • slight increase in blood pressure

Less common reactions:

  • dizziness
  • fatigue
  • darkening of the skin with prolonged use

In most cases, these effects are short-lived and appear soon after administration, especially during the adaptation process.

Comparison of PT-141 (Bremelanotide) with other peptides

PT-141 belongs to the group of melanocortin agonists, but it stands out with a strongly expressed central effect and a focus on sexual function.

PT-141 vs Melanotan II
Both peptides originate from the same group, but Melanotan II has a broader effect on the melanocortin system, including pigmentation, appetite, and libido. PT-141 is more selective and optimised mainly for sexual function, with a weaker influence on skin pigmentation.

PT-141 vs Oxytocin
Oxytocin affects social bonding and emotional closeness, while PT-141 directly activates sexual desire. The difference is that Oxytocin works on the emotional aspect, while PT-141 works on the arousal mechanism itself.

PT-141 vs Kisspeptin

Kisspeptin works at a hormonal level by activating the HPG axis and increasing GnRH and testosterone. In short, PT-141 provides a direct neurological effect on libido, while Kisspeptin regulates the environment that controls it.
