What is Retatrutide?

Retatrutide is a new generation synthetic peptide, developed as a triple receptor agonist that simultaneously activates GLP-1, GIP and glucagon receptors. This makes it one of the most innovative molecules in current research on metabolism and body composition.

The peptide was initially created by the pharmaceutical company Eli Lilly with the aim of enhancing the effects of classic GLP-1 therapies through combined action on several key hormonal pathways. Unlike standard solutions that target only appetite or glucose, Retatrutide simultaneously affects energy intake, energy expenditure and metabolic regulation.

The GLP-1 component suppresses appetite and delays gastric emptying, GIP improves insulin sensitivity, and the glucagon effect increases energy expenditure. It is this combination that makes Retatrutide significantly more powerful compared to previous generations of peptides.

Development of Retatrutide began around 2018–2019, and currently the peptide is in late-stage clinical trials and is considered one of the most promising future therapies for obesity and metabolic syndrome.

The results show a clear dose-dependent effect, with higher doses leading to more pronounced weight reduction.

Retatrutide is currently in phase III clinical trials, which will determine its final profile and possible approval for medical use.

Proper administration and dosage of Retatrutide within a research protocol

Retatrutide is administered by subcutaneous injection once weekly, with the main principle being gradual and controlled dose escalation to achieve optimal tolerability and minimize side effects.

In most research protocols, treatment starts with an initial dose of about 1 – 2 mg weekly, after which the dose is increased stepwise every 4 weeks up to 4 mg. With good tolerability and depending on the goals of the protocol, it may be advanced to higher doses of 6 mg to 8 mg weekly.

This type of titration allows gradual adaptation to the peptide's combined mechanism, which simultaneously affects appetite, energy expenditure and metabolic regulation.

The duration of the protocol usually ranges between 12 and 24 weeks, and in longer-term studies a cyclical approach with rest periods is often used to assess the effect and restore sensitivity.

Gradual dose escalation is a key factor as it significantly reduces the risk of gastrointestinal reactions and improves overall tolerability to the peptide.

Possible side effects of Retatrutide

As with other peptides in this class, side effects are most often related to the gastrointestinal system and are dose-dependent.

Retatrutide is often compared to Semaglutide and Tirzepatide because all three molecules are among the most potent modern peptides for weight control and metabolic disorders. The difference is that Semaglutide acts as a GLP-1 receptor agonist, Tirzepatide is a dual agonist of GIP and GLP-1, and Retatrutide is a triple agonist that activates GLP-1, GIP and glucagon receptors. It is this triple activation that is why Retatrutide is considered a next-generation metabolic peptide.

Compared to Semaglutide, Retatrutide has a broader mechanism of action. Semaglutide is an excellent choice for reducing appetite, improving glycemic control and weight reduction, being established in obesity treatment at doses up to 2.4 mg weekly. In Semaglutide clinical trials weight reduction is usually around 15 percent in longer research protocols, which makes it extremely effective, but still more limited in metabolic scope compared to Retatrutide.

Compared to Tirzepatide, Retatrutide again has an important step forward. Tirzepatide combines GIP and GLP-1 and is significantly stronger than classic Semaglutide, being approved for obesity and overweight and in practice used at doses up to 15 mg weekly. It shows very strong results in weight reduction, but Retatrutide adds a third mechanism via the glucagon receptor, which provides potential for higher energy expenditure and a more aggressive impact on adipose tissue.

In terms of efficacy, available clinical data place Retatrutide at the top of this category. In phase II studies Retatrutide shows reductions in body weight of up to around 24 percent, while Tirzepatide usually performs lower, and Semaglutide remains another step behind. This does not mean the other two peptides are weak, but that Retatrutide currently appears to be the most potent molecule in this class.

In short, Semaglutide is an excellent choice when a proven and stable GLP-1 effect is sought, Tirzepatide is the next level with a stronger effect on weight and glucose, and Retatrutide is considered the most complex and most aggressive option for research on body composition and metabolic control.
